What are Single-Domain Antibodies?

Single-domain antibodies (also known as VHH) are small, ~15kDa fragments isolated from a special type of heavy chain-only antibody that is produced by camelids, such as llamas and alpacas.

Our vertically integrated single-domain antibody discovery process begins with immunization of llamas and alpacas. Proprietary methods are used to ensure the VHH region is amplified and cloned into the phagemid for phage display panning and screening for VHH hits, resulting in complete sequences of your custom VHH library or recombinant expression of the single-domain antibody.

High Quality Cells and RNA:

We strive to preserve cell health for RNA isolation. Animal blood is processed quickly to purify peripheral mononuclear cells. RNA is double-purified with phenol-chloroform extraction and spin columns and examined by electrophoresis prior to making cDNA to ensure cell quality meets our standards before generation of the cDNA library.

Unique PCR Primers:

We perform extensive bioinformatics research on VHH genes and design novel primers for RT-PCR to ensure we amplify the VHH cDNA from camelid antibodies and not conventional antibodies. VHH libraries constructed with these primers have higher diversity and larger coverage of VHH repertoires.

Robust Independent Clones:

We deliver libraries with large numbers of independent clones. Our PBMC isolation and library construction protocol routinely yields >1×108 PBMC cells from each production bleed.
