Antibodies: The Key to Spatial Mass Spectrometry

As multiplex immunofluorescence techniques like imaging mass cytometry (IMC) gain widespread adoption, assay performance increasingly depends on antibody quality and validation. Unlike traditional methods, IMC uses metal-labeled antibodies and enables simultaneous application of all primaries, but results can be compromised by reduced antibody activity following conjugation or insufficient optimization. 


This study investigates whether systematic evaluation of antibody performance both before and after metal conjugation can serve as a reliable predictor of IMC success, offering a practical strategy to improve assay consistency and data quality.
