Antibody was affinity purified using an epitope specific to IRS1 immobilized on solid support.

The epitope recognized by IHC-00357 maps to a region between residue 1192 and 1242 of human insulin receptor substrate 1 using the numbering given in entry NP_005535.1 (GeneID 3667).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

The insulin receptor substrate 1 (IRS1) is one of four members of the IRS family (IRS1-4) that mediates intracellular signaling downstream of the IGF1 and insulin receptors. Phosphorylation of IRS1 by activated receptors leads to binding of IRS to the SH2 domains of effector proteins such as PI3K, Grb2, nck, and SHP2. The downstream effects of insulin and IGF1 signaling via IRSs on the cell are pleiotropic and include various responses such as glucose transport, glycogen synthesis, protein synthesis, antilipolysis, and gluconeogenesis. Polymorphisms in IRS1 may contribute to the etiology of non-insulin-dependent diabetes mellitus (NIDDM).
