The SMARCs (SWI/SNF-related, matrix-associated, actin-dependent regulators of chromatin), and BAFs (BRG1-associated factors), have been identified as components of the mammalian SWI/SNF-like chromatin-remodeling protein complexes. These multi-protein complexes are proposed to function as ATP-driven motors that translocate along DNA and destabilize nucleosomal structures to facilitate transcription factor binding. SMARCD1/BAF60a is subunit of the SWI/SNF complex that has been identified as a factor that interacts with nuclear receptors such as the vitamin D receptor and glucocorticoid receptor and a subset of Fos/Jun dimers. SMARCD1 has also been shown to interact with p53 and is suggested to be involved in the tumor suppressor activity of p53. From these findings it has been concluded that SMARCD1 functions as a direct link between transcription factors and SWI/SNF remodeling activity to regulate transcription.
