Antibody was affinity purified using an epitope specific to UBR1 immobilized on solid support.

The epitope recognized by A302-988A maps to a region between residue 1125 and 1175 of human Ubiquitin Protein Ligase E3 Component n-Recognin 1 using the numbering given in entry NP_777576.1 (GeneID 197131).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

UBR1 is an E3 ubiquitin-protein ligase that is a component of the N-end rule pathway. UBR1 recognizes and binds to proteins bearing specific N-terminal residues that are destabilizing according to the N-end rule, leading to their ubiquitination and subsequent degradation. UBR1 may be involved in pancreatic homeostasis. UBR1 binds leucine and is a negative regulator of the leucine-mTOR signaling pathway, thereby controlling cell growth [taken from the Universal Protein Resource (UniProt) www.uniprot.org/uniprot/ Q8IWV7].

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
