Anti-T-Cell Immunization Strategy Yields New AbNano VHH Library

To advance targeted antibody discovery against complex antigens on the surface of T-Cells, we developed the AbNano™ Anti-T-cell VHH library.

Our immune-derived library began with immunizing unstimulated, CD3+ human T-Cells and isolating approximately one million unique VHH sequences. This preliminary pool was further diversified through serial error-prone PCR, and transformation yielded up to one billion unique VHH molecules at the protein level. This extensive diversity enables rapid discovery of nanomolar-affinity binders to T-cell markers like CD3ε and CD8.

The poster presents quantitative diversity analysis, hit rates, and affinity data, including dozens of validated binders identified by ELISA and NGS with clear illustrations of each step.

Unlike synthetic libraries, AbNano™ VHH libraries deliver immune-driven diversity for more relevant antibody discovery. This modular workflow is adaptable for future antibody campaigns in immunology and immuno-oncology.

Screening Results:

  • For CD3ε: 34 unique CDR3 sequences from 116 hits.
  • For CD8: 43 unique CDR3 sequences from 58 hits.
  • Affinity: High nanomolar to low micromolar EC50s for selected clones.
