Product Description

ErbB2 is a member of the receptor tyrosine kinase family of proteins. The four members of this family are ErbB1 (Epidermal growth factor receptor; EGFR/HER1), ErbB2 (HER2/Neu), ErbB3 (HER3), and ErbB4 (HER4). The ErbB receptor members are single-pass transmembrane proteins with an extracellular ligand-binding domain and a cytoplasmic tyrosine-kinase domain. The epidermal growth factor (EGF) and its related proteins serve as the ligands for the ErbB receptors. Upon binding, the receptors heterodimerize, autophosphorylate and make contact with adapter molecules to transmit cellular signals involved in cell proliferation, survival, and differentiation. The ErbB2 receptor is the only member that does not interact with a ligand, but instead functions as a heterodimeric partner to coordinate signals from other ligand bound ErbB receptors. Amplification and/or overexpression of ErbB2 have been reported in many cancers.

Production & Epitope

Antibody was affinity purified using an epitope specific to ErbB2 immobilized on solid support.

The epitope recognized by A300-621A maps to a region between residues 975 and 1025 of human v-erb-b2 erythroblastic leukemia viral oncogene homolog 2 using the numbering given in entry NP_004439.2 (GeneID 2064).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).

Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.

Immunohistochemistry (IHC)1:100 - 1:500. Epitope retrieval with Tris-EDTA pH 9.0 is recommended for FFPE tissue sections.
Immunoprecipitation (IP)6 µg/mg lysate
Western Blot (WB)1:2000 - 1:10,000
