Product Description

The gene EPAS1 encodes for the transcription factor endothelial PAS domain-containing protein 1, and is induced as oxygen levels fall. Commonly called hypoxia inducible factor 2A (HIF2a), this protein is involved in the induction of oxygen regulated genes, binding to core DNA sequence 5'-[AG]CGTG-3' within the hypoxia response element (HRE) of target gene promoters. An important cancer biomarker, HIF2a may also play a role in Tibetan high-altitude adaptation (Am J Hum Genet. 2015 Jul 2; 97(1): 54–66.) and mutations in the EPAS1 gene are associated with erythrocytosis familial type 4 [taken from NCBI Entrez Gene (Gene ID: 2034) and UniProtKB (Entry: Q99814)].

Production & Epitope

Recombinant antibody was purified from cell culture supernatant.

The epitope recognized by A700-003 maps to a region between residues 400 and 450 of human endothelial PAS domain protein 1 using the numbering given in entry NP_001421.2 (GeneID 2034).

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).

Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.

ChIP-SeqA previous lot has qualified for ChIP-Seq. 10 - 40 µl per 30 µg chromatin.
Immunocytochemistry (ICC)1:200 - 1:1000. Formaldehyde fixation is recommended. Permeabilization with Triton-X 100 is recommended for formaldehyde-fixed cells.
