Product Description

DHX9 is a member of the DEAH box family of proteins that possesses several conserved motifs which include the highly conserved DEAH (Asp-Glu-Ala-His) amino acid sequence motif. The major activity of DEAH box proteins is to function as ATP-dependent RNA helicases. RNA helicases play an important role in all aspects of RNA metabolism and function which include pre-mRNA splicing, RNA synthesis, RNA degradation, RNA export, RNA translation, RNA secondary structure formation, ribosome biogenesis, and the assembly of RNP complexes. Some members of the DEAH box proteins also exhibit functions involved in transcriptional regulation. DHX9 has been shown to act as a bridge between CREB-binding protein (CBP) and RNA polymerase II to coactivate CBP-dependent transcription. A similar observation has been reported for an interaction with BRCA1. DHX9 also exhibits activities associated with nuclear export of retroviral RNAs, translation, RNA editing, and snRNP assembly.

Production & Epitope

Antibody was affinity purified using an epitope specific to DHX9 immobilized on solid support.

The epitope recognized by A300-855A maps to a region between residue 1220 and the C-terminus (residue 1270) of human DEAH (Asp-Glu-Ala-His) Box Polypeptide 9 using the numbering given in entry CAA71668.1 (GeneID 1660).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).

Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.

Immunohistochemistry (IHC)1:100 - 1:500. Epitope retrieval with citrate buffer pH6.0 is recommended for FFPE tissue sections.
Immunoprecipitation (IP)6 µg/mg lysate
Western Blot (WB)1:2000 - 1:10,000
