Product Description

Splicing factor 45 (SPF45) is an RNA splicing factor that binds to the single stranded 3'AG at the exon/intron border and promotes its utilization in the second catalytic step. SPF45 is involved in the regulation of alternative splicing and the utilization of cryptic splice sites. It has been shown to promote the utilization of a cryptic splice site created by the beta-110 mutation in the human beta-thalassemia gene. The resulting frameshift leads to sickle cell anemia [taken from the Universal Protein Resource (UniProt) www.uniprot.org/uniprot/Q96125].

Production & Epitope

Antibody was affinity purified using an epitope specific to SPF45 immobilized on solid support.

The epitope recognized by A302-497A maps to a region between residue 25 and 75 of human splicing factor (45kD) using the numbering given in entry NP_116294.1 (GeneID 84991).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.

Immunohistochemistry (IHC)1:1000 - 1:5000. Epitope retrieval with citrate buffer pH6.0 is recommended for FFPE tissue sections.
Immunoprecipitation (IP)6 µg/mg lysate
