Product Description

AKAP8/AKAP95 has been characterized as a 95 kDa protein kinase A (PKA)-anchoring protein and regulatory subunit of PKA that functions to recruit the condensin complex for chromosome condensation during mitosis. A role in DNA replication has also been demonstrated through its interaction with the pre-replication complex component, MCM2 (mini-chromosome maintenance 2). Inhibition of AKAP8/AKAP95 association with MCM2 inhibits replication initiation and elongation in vitro. AKAP8/AKAP95 has also been shown to be involved in apoptosis. KAP8/AKAP95 associates with caspase 3 and serves as a carrier to transport caspase 3 from the cytoplasm to the nucleus to induce morphological changes in the nucleus.

Production & Epitope

Antibody was affinity purified using an epitope specific to AKAP8/AKAP95 immobilized on solid support.

The epitope recognized by IHC-00217 maps to a region between residue 642 and the C-terminus (residue 692) of human A-kinase anchor protein 8 using the numbering given in entry NP_005849.1 (GeneID 10270).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

Epitope exposure is recommended.

Epitope exposure with citrate buffer will enhance staining.

Likely to work with frozen sections.

In some cases, the antibody may be diluted further than indicated.

Immunohistochemistry (IHC)1:100 - 1:500
