Product Description

LARG (leukemia-associated RhoGEF) is homologous to PDZ-RhoGEF and functions as a RhoA-specific guanine nucleotide exchange factor. RhoGEFs catalyze the exchange of GDP for GTP on the GTPase, Rho, to activate downstream signaling pathways. LARG contains a PDZ [post synaptic density protein (PSD95), Drosophila discs-large tumor suppressor (DIgA), and zonula occludens-1 protein (zo-1)] domain which is an 80-90 amino acid structural domain found in signaling proteins in bacteria, yeast, plants, and animals. LARG is proposed to play a role in cell polarity and microtubule dynamics and may act as a GTPase-activating protein (GAP) for the heterotrimeric Galpha(12) and Galpha(13) proteins.

Production & Epitope

Antibody was affinity purified using an epitope specific to LARG immobilized on solid support.

The epitope recognized by A301-960A maps to a region between residue 1494 and 1544 of human leukemia-associated rho guanine nucleotide exchange factor using the numbering given in entry NP_056128.1 (GeneID 23365).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).

Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.

Immunoprecipitation (IP)6 µg/mg lysate
Western Blot (WB)1:2000 - 1:10,000
