Antibody was affinity purified using an epitope specific to p66alpha immobilized on solid support.

The epitope recognized by A302-358A maps to a region between residue 573 and 633 of human p66alpha using the numbering given in entry AAO31797.1 (GeneID 54815).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

p66alpha and p66beta are human orthologs of the xenopus p66 subunit of the Mi-2/NuRD complex. The Mi-2/NuRD complex is a multi-subunit complex that mediates gene repression via processes involving chromatin remodeling and histone deacetylation. p66 alpha and beta were identified in a two-hybrid screen using the methyl-CpG-binding domain protein, MBD2, as bait. It has been demonstrated that p66alpha and p66beta mediate the interaction between MBD2 and histones.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
