Product Description
Chk1 functions as a checkpoint protein kinase that is critical to the regulation of the Cdc25a and Cdc25c phosphatases that promote S and G2/M phase progression, respectively. Chk1 kinase activity inhibits cell cycle progression in the presence of DNA damage by phosphorylating Cdc25a and Cdc25c and causing the degradation of Cdc25a and cytoplasmic sequestration of Cdc25c.

Antibody was affinity purified using an epitope specific to Chk1, Phospho (S317) immobilized on solid support.
Immunogen for A304-673A was a phosphorylated synthetic peptide, which represented a portion of human Checkpoint Kinase 1 (GeneID 1111) around serine 317 according to the numbering given in entry NP_001265.1. of human Checkpoint Kinase 1 using the numbering given in entry NP_001265.1 (GeneID 1111).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.
Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).
Immunohistochemistry (IHC)1:500 to 1:2,000. Epitope retrieval with citrate buffer pH6.0 is recommended for FFPE tissue sections.
Immunocytochemistry (ICC)1:500 to 1:2,000. Epitope retrieval with citrate buffer pH6.0 is recommended for FFPE cell sections.
