Product Description

Rad17 is a cell cycle checkpoint gene required for cell cycle arrest and DNA damage repair in response to DNA damage. This protein shares strong similarity with DNA replication factor C (RFC), and can form a complex with RFCs. This protein binds to chromatin prior to DNA damage and is phosphorylated by the checkpoint kinase ATR following damage. This protein recruits the RAD1-RAD9-HUS1 checkpoint protein complex onto chromatin after DNA damage, which may be required for its phosphorylation. The phosphorylation of this protein is required for the DNA-damage-induced cell cycle G2 arrest, and is thought to be a critical early event during checkpoint signaling in DNA-damaged cells. Production & Epitope

Antibody was affinity purified using a synthetic peptide representing phosphorylation at Serine 645 and surrounding residues of Rad17 immobilized on solid support.

Immunogen for A300-153A was a phosphorylated synthetic peptide, which represented a portion of human Rad17 (GeneID 5884) around serine 645 according to the numbering given in entry NP_002864.1.

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).

Immunohistochemistry (IHC)1:500 - 1:2000. Epitope retrieval with citrate buffer pH 6.0 is recommended for FFPE tissue sections.
Western Blot (WB)1:1000 - 1:5000
