The epitope recognized by A300-731A maps to a region between residue 1 and 50 of human Protein Phosphatase 2A (PPP2CB, Protein Phosphatase 2, Catalytic Subunit, Beta isoform) using the numbering given in entry NP_004147.1 (GeneID 5516). The region is highly conserved with PPP2CA (NP_002706.1; GeneID: 5515)
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to PP2A immobilized on solid support.

PP2A is a major serine/threonine phosphatase that is implicated in the negative control of cell cycle progression. PP2A encodes the beta isoform of the catalytic subunit (PP2CB). Chk2 has been shown to be negatively regulated and dephosphorylated by PP2A. Several proteins have been shown to associate with PP2A and regulate its activity such as eRF1, Hox11, CK2, and Igbp1/alpha4. Rapamycin has been shown to dissociate PP2A from alpha-4 and inhibit proliferation. This result suggests an alternative positive regulatory role of PP2A in cell proliferation.

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
