Antibody was affinity purified using an epitope specific to RFC1 immobilized on solid support.
The epitope recognized by A300-320A maps to a region between residue 1100 and the C-terminus (residue 1148) of human Replication Factor C subunit 1 using the numbering given in entry NP_002904.3 (GeneID 5981).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

The Replication factor C (RFC) heteropentamer includes RFC1, RFC2, RFC3, RFC4, and RFC5. The RFC heteropentamer is part of the multiprotein DNA replicase that functions to replicate DNA prior to cell division and repair DNA damage. RFC is the clamp loader ATPase associated with the DNA replicase. As the clamp loader, RFC uses ATP to recruit, open, and close the PCNA ring to link it to DNA for replication and repair. RFC1 is the 140 kDa subunit of RFC that has a DNA-binding domain with a BRCT motif and is suggested to play a more critical role than the other subunits.
