Production Procedures

Antibody was affinity purified using an epitope specific to UACA immobilized on solid support.

The epitope recognized by A301-382A maps to a region between residue 675 and 725 of human uveal autoantigen with coiled-coil domains and ankyrin repeats using the numbering given in entry NP_060473.2 (GeneID 55075).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Additional Product Information

UACA (uveal autoantigen with coiled-coil domains and ankyrin repeats) was originally identified as an autoantigen in patients with panuveitis, a characteristic of Vogt-Koyanagi-Harada disease, and in patients with Graves’ disease. UACA was also later identified as Nucling, an mRNA differentially expressed in F9 embryonal carcinoma cells during cardiac muscle differentiation. UACA appears to function as a pro-apoptotic protein that recruits the apaf-1-pro-caspase-9 complex for the induction of apoptosis to mediate the cell-death pathway.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
