Product Description

MEK1 is an activator and kinase for MAP and ERK kinases. It is a serine/threonine kinase essential to the MAP kinase signal transduction pathway which is involved in a multitude of cellular processes such as proliferation, differentiation, gene activation, and development. MEK1 displays 85% identity with MEK2. Although MEK1 and MEK2 have been found to be functionally similar, they may serve non-redundant functions. Both are required for cellular proliferation and may play a role in tumorigenesis.

Production & Epitope

Antibody was affinity purified using an epitope specific to Phospho MEK1 immobilized on solid support.

The epitope recognized by A303-610A maps to a region of human MAPK/ERK kinase 1 (dual specificity mitogen-activiated protein kinase kinase 1) surrounding threonine 286 using the numbering given in entry NP_002746.1 (Gene ID 5604) when the threonine residue is phosphorylated.

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.

Immunohistochemistry (IHC)1:1000 - 1:5000. Epitope retrieval with citrate buffer pH 6.0 is recommended for FFPE tissue sections.
Immunoprecipitation (IP)6 µg/mg lysate
