Product Description
Bromodomain-containing protein 4 (BRD4) contains 2 bromo-domains. Bromo-domains are found in chromatin-associating proteins and can interact specifically with acetylated lysine. BRD4 is described as a chromatin adaptor and has been shown to bind acetylated chromatin. It was originally identified as MCAP (mitotic chromosome associated protein) due to its association with mitotic chromosomes. BRD4 also associates with several transcription complexes such as the Mediator complexes and P-TEFb complexes and is considered to function as a transcriptional co-factor.

Recombinant antibody was purified from cell culture supernatant.
The epitope recognized by A700-004 maps to a region between residue 1312 and 1362 of human bromodomain-containing protein 4 using the numbering given in entry NP_490597.1 (GeneID 23476).
Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (A120-101P).
Western blots of immunoprecipitates are performed using an anti-Rabbit IgG Light Chain HRP-conjugated antibody with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
ChIP-SeqA previous lot has qualified for ChIP-Seq. 10 - 40 µl per 30 µg chromatin.
Flow Cytometry (Flow Cyt)Fixed in 4% formaldehyde and permeabilized with 90% methanol. 1 µl per 1 x 10^6 cells.
Immunocytochemistry (ICC)1:100 to 1:500. Epitope retrieval with citrate buffer pH6.0 is recommended for FFPE cell sections.
Immunocytochemistry-Immunofluoresence (ICC-IF)1:25 - 1:250. Formaldehyde fixation is recommended. Permeabilization with Triton-X 100 is recommended for formaldehyde-fixed cells.
Immunohistochemistry (IHC)1:100 to 1:500. Epitope retrieval with citrate buffer pH6.0 is recommended for FFPE tissue sections.
