Production Procedures
Antiserum was solid phase adsorbed to ensure subclass specificity. Antiserum was cross adsorbed using human and rat immunosorbents to remove cross reactive antibodies. The antibody to mouse IgG3 was isolated by affinity chromatography using antigen coupled to agarose beads and conjugated to fluorescein isothiocyanate (FITC).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.
By immunoelectrophoresis and ELISA this antibody reacts specifically with mouse IgG3. Cross reactivity with other immunoglobulins is less than 2%. No antibody was detected against non-immunoglobulin serum proteins. Less than 2% cross reactivity to human and rat IgG3 was detected. This antibody may cross react with IgG3 from other species.

IgG3 is a minor class of IgG found in the serum, but has a unique hinge region and contains more glycosylation sites than other IgG molecules, which make increase its potency within the immune system.
